- Some patients have large increases in triglycerides and cholesterol. The long-term chance of getting complications such as heart attacks or stroke due to increases in triglycerides and cholesterol caused by protease inhibitors is not known at this time.
- Diabetes and high blood sugar (hyperglycemia) occur in patients taking protease inhibitors such as this medicine. Some patients had diabetes before starting protease inhibitors, others did not. Some patients need changes in their diabetes medicine. Others needed new diabetes medicine.
- Changes in body fat have been seen in some patients taking antiretroviral therapy. These changes may include increased amount of fat in the upper back and neck ("buffalo hump"), breast, and around the trunk. Loss of fat from the legs, arms and face may also happen. The cause and long term health effects of these conditions are not known at this time.
- Some patients with hemophilia have increased bleeding with protease inhibitors. There have been other side effects in patients taking this medicine. Shingles

For preventing future outbreaks of genital herpes, the recommended dose is Valtrex 1000 mg (1 gram), taken once a day, every day. For people who have had fewer than ten outbreaks of genital herpes in one year, the Valtrex dosage may be decreased to 500 mg a day, every day.
For people with HIV and genital herpes who have a CD4 cell count equal to or greater than 100 cells/mm3, the dose of Valtrex to prevent future outbreaks is 500 mg, taken twice a day, every day. Valtrex has not been studied in people with HIV who have a CD4 cell count less than 100 cells/mm3.

Kidney failure and nervous system problems have happened in patients who already have kidney disease and in elderly patients whose kidneys do not work well due to age. The most common side effect seen in children <18 years of age is headache.

- In patients receiving a potent CYP3A4 inducer such as rifampicin or phenytoin, a dose increase to 500 mg daily should be considered in the absence of severe adverse drug reactions, and clinical response and adverse events should be carefully monitored.
- No dosage adjustment is required on the basis of patient age, body weight, gender, ethnicity or renal function, or in patients with moderate-to-severe hepatic impairment due to liver metastases.
Paediatric Population

Hepatic Impairment
Patients with moderate-to-severe hepatic impairment (Child-Pugh B or C) due to cirrhosis have increased plasma concentrations of gefitinib. These patients should be closely monitored for adverse events. Plasma concentrations were not increased in patients with elevated aspartate transaminase (AST), alkaline phosphatase or bilirubin due to liver metastases.
Renal Impairment
